Oligonucleotide therapeutics have delivered on their promise in the liver, but the field's next frontier lies elsewhere. Naked and chemically stabilized oligonucleotides rely on passive uptake and accumulate predominantly in the liver and kidney regardless of the intended target, leaving much of the disease landscape out of reach. Conjugation is emerging as the central strategy for changing that, extending delivery to tissues that were previously inaccessible.
This collaborative panel between Nucleic Acid Insights and Bioconjugate Insights brings together experts from both the oligonucleotide and bioconjugate communities to examine how conjugation is redrawing the map of oligonucleotide delivery, tracing conjugate-enabled targeting across tissues, from antibody-based delivery already in the clinic for muscle through active development in the central nervous system and other extrahepatic targets.
